Yang et al (Immunity 2024 Apr 5:S1074-7613(24)00138-9; https://doi.org/10.1016/j.immuni.2024.03.017) uncovered a novel function for intraepithelial mast cells in gasdermin activation. Upon helminth an infection, intraepithelial mast cell-derived proteases activated the plasma membrane pore-forming enzyme gasdermin C inside intestinal enterocytes with subsequent launch of the potent sort 2 cytokine IL-33. Notably, each epithelium-intrinsic gasdermin C activation and IL-33-mediated sort 2 immune responses had been abolished with mast cell deficiency.
